John's wort, carbamazepine): May decrease concentrations of sofosbuvir and/or velpatasvir. ( 7.3 ) 7.1 Potential for Other Drugs to Affect EPCLUSA Sofosbuvir and velpatasvir are substrates of drug transporters P-gp and BCRP while GS-331007 (the predominant circulating metabolite of sofosbuvir) is not. In vitro, slow metabolic turnover of velpatasvir by CYP2B6, CYP2C8, and CYP3A4 was observed.
